An iSeq v2 reagent cartridge (20031371, Illumina) is thawed in a water bath at room temperature for 1?h; the flow cell is inserted; and the kit is kept at room temperature. The LAMP-Seq library and the N-100 random-index library are diluted to 2?nM each in water. Next, 1?µl of the combined library is mixed with 150?µl of Tris pH 8.5. Then, 20?µl of the final dilution is loaded into the reagent cartridge and sequenced according to the manufacturer’s instruction using the following cycle numbers:
